چکیده

AP-2 is a family of transcription factors that play important roles during embryonic development. Two AP-2 genes, AP-2 and AP-2 , have previously been characterized in chick retina. Here, we demonstrate that a third member of the chicken AP-2 family, AP-2 , is primarily expressed in the retina and brain, with highest levels at embryonic days 7 to 11. By in situ hybridization and immunohistochemical analysis, we show that AP-2 RNA and protein are found in a subset of ganglion cells in embryonic chick retina. Coimmunostaining with anti-Brn3a and anti–AP-2 antibodies indicates that approximately one-third of Brn3a-positive ganglion cells express AP-2 . AP-2 RNA but not AP-2 protein is also found in cells located in the outer half of the inner nuclear layer. The spatial and temporal distribution of AP-2 protein in the retina suggests a transient role in a subset of late-born ganglion cells likely involving axonal trafficking or pathfinding. Differential expression of AP-2alpha and AP-2beta in the developing chick retina: repression of R-FABP promoter activity by AP-2.

Retinal fatty acid binding protein (R-FABP) is the avian counterpart of murine brain FABP implicated in glial cell differentiation and neuronal cell migration. R-FABP is highly expressed in the undifferentiated retina and brain of chick embryos. We have previously shown by in vitro studies that the AP-2 transcription factor binds to a consensus AP-2 binding site in the R-FABP promoter region. B...

Quantitative Analysis of GABAA Gamma Receptor Subunits in the Developing Embryonic Chick Forebrain

Objective(s) In this study we investigated the expression of GABAA receptor subunits during brain development. These receptors may change in the embryonic chick forebrain. Materials and Methodes The expression levels of four types of GABAA receptor gamma subunits (γ1, γ2, γ3 and γ4) were quantified in the embryonic chick forebrain at 32 hr, 3, 7, 14, and 20 days of incubation and day one aft...

Transcription factor AP-2delta regulates the expression of polysialyltransferase ST8SIA2 in chick retina.

The AP-2δ transcription factor is restricted to a subset of retinal ganglion cells. Overexpression of AP-2δ in chick retina results in induction of polysialylated neural cell adhesion molecule (PSA-NCAM) accompanied by misrouting and bundling of ganglion cell axons. Two polysialyltransferases, ST8SIA2 and ST8SIA4, are responsible for polysialylation of NCAM. Here, we investigate the mechanism d...

The 2M6 antigen is a Müller cell-specific intracellular membrane-associated protein of the sarcolemmal-membrane-associated protein family and is also TopAP

PURPOSE The differentiation marker 2M6 has been used to identify Müller cells within the developing chick retina for several years, although the molecular identity of 2M6 was not known. This study was aimed at determining the identity of the protein antigen recognized by the 2M6 monoclonal antibody. METHODS Affinity chromatography and subsequent mass spectrometry were used to determine the mo...

Involvement of AP-2 in regulation of the R-FABP gene in the developing chick retina.

Little is known regarding the molecular pathways that underlie the retinal maturation process. We are studying the regulation of the retinal fatty-acid-binding protein (R-FABP) gene, highly expressed in retinal precursor cells, to identify DNA regulatory elements and transcriptional factors involved in retinal development. Although the upstream sequence of the R-FABP gene is extremely GC rich, ...
